Cloning and Sequence Analysis of the Polygalacturonase (PG) Gene in Pepper (Capsicum Annuum L.)
Cv. P98 of Pepper (Capsicum annuum L.) was used in this study. A full-length PG cDNA was isolated by rapid amplification of cDNA ends (RACE) and PCR, which designated as CaPG (GenBank accession number: FJ596175). The full-length CaPG gene contained a complete open reading frame (ORF) of 1107 bp, which encoded a 369 amino acids polypeptide. Amino acid sequence homo logy analysis of CaPG showed an 84% identity with PG-2a in tomato and 85% identity with PG in papaya. The result of phylogenetic analysis showed that PG gene from pepper and from tomato and papaya clustered together, and then came those from Arabidopsis.
