Cloning and Expression of WRKY Gene Segment in Celery
The WRKY transcription factor is considered to be involved in biotic and abiotic stress responses widely in plant. In this study, we acquired a 682 bp WRKY segment and a 1005 bp β -actin segment, induced WRKY gene expression in celery seedlings under the following treatment conditions: salinity cold (4℃), (200 mM NaCI), and abscisic acid (ABA) (100 μM), and finally detected different expressions of WRKY gene by using RT-qPCR (real-time relative quantification PCR). The results of RT-qPCR analysis revealed that the relative expression of WRKY gene improved 256.93%, 207.19%, 1825.37% after cold, salinity, and ABA treatment respectively, which demonstrated that WRKY gene plays a positive regulatory role under different conditions.
