Lilium Pumilum Bulblets Formation in vitro: Effects of Plant Growth Regulators and Culture Condition

The second to fifth outer scales of Lilium pumilum bulb were selected as explants. The results shcnv that the optimum combination for callus induction is MS medium with the addition of 1.0 mg/L BA and 0.6 mg/L NAA, and the best medium for the multiplication of adventitious buds is MS supplemented with 1.0 mg/L BA and 0.1 mg/L NAA. The inductivity of both callus and adventitious buds are higher significantly in basal part than which in the middle and top of scale. SA, AC as well as PP333 added in regeneration media are advantageous both for the formation and development of bulblets in vitro. Illumination can help improve the propagation coefficient, and especially in the medium added 1.0 μM SA, the highest proliferous rate is obtained. By contrast, dark condition can obviously increase bulblet diameter and get the best breeding individuals, and 5.0 mg/L PP333 is the best additives for bulblets growing.
