Comparative Investigation on in Vitro Regeneration of Different Rhubarb Cultivars
Differences in regeneration ability of five rhubarb cultivars were investigated using shoot tips of in vitro seedling on MS medium supplemented with 1.0mg/L NAA and 1.0mg/L 6-BA. Two different patterns of shoot proliferation were noted after 24 days of culture, i.e. caespitose shoots proliferating from callus of explants and axillary shoot bourgeoning from explants. A significant difference in regenerative ability was observed, with cultivar RT showing the highest multiplication coefficient (3.71/explant) and R19 presenting the lowest multiplication coefficient (1.96/explant). The result suggested that subculture for multiplication can be conducted in every 24 days to establish a micro-propagation system.
