Break of the Dormancy of Malus hupehensis Rehd. Embryo by Low Temperature and Plant Growth Regulators in vitro

The embryos of Malus hupehensis Rehd. were used for studying the influence of low temperature and plant growth regulators such as 6-BA and GA3 on breaking dormancy in vitro. The results indicated that low temperature had significant effects on breaking dormancy, and the lower temperature and longer time of treatment was beneficial to the seedling rate. When the embryos of Malus hupehensis Rehd. which had no low temperature treatment were cultured on the MS medium supplemented with plant growth regulators, the dormancy could be abolished, especially in MS+GA32.0mg/L+6-BA0.2mg/L the results were the best and the percent of plant formation was 94.1%. Combination of low temperature and plant growth regulators showed the best effect on the breaking dormancy.
