Rescue of Hybrid Embryos Before Their Aborting in Earlier Stage in Parthenocarpic Mopanshi Persimmon by An Ovule Culture Procedure
There were two kinds of abortive phenomena observed in crosses with Chinese parthenocarpic Mopanshi persimmon as female parent The abortion began at 35 days after pollination (DAP) and 60 DAP, respectively. In this report, an ovule culture procedure was developed to rescue globular zygotic embryos that were to abort in the earlier stage (35 DAP). The optimum medium supplements in ovule and/or embryo culture stage were determined and evaluated by using L8 (4X24) orthogonal design when a modified solid MS (half of NO3 in Murashige and Skoog) was used as the base medium. The successful procedure was established as follows: the fertilized ovules excised at 35 DAP were cultured in the appropriate medium for ovule developing MS(1/2NO3)+ IAA 11.42|iM+ BA 2.22uM+ GA 1.44uM +7V. sucrose for initial 15 days , then the obtained heart-shaped embryos were transferred to the (determined)embryo developing medium MS(1/2NO3)+ IAA 2.85uM+ BA 4.44uM+ 7% sucrose| for 20 days followed by another 20 day subculture in embryo germinating medium(MS (3/4NO3 )+ IBA 0.1uM+ BA 0.44uM+ 5%siicrose). The results showed that this technique could eventually succeed in rescuing 49% ovules with globular embryos developed to plantlets and thus might be a desirable approach to obtain hybrids of persimmon in breeding practice restricted by similar post-fertilization barriers.
