Impact of a-naphthalene Acetic Ccid on Peony Embryo Culture
Using the mature embryo of the tree peony cultivar viz. Fengdan as the explants, the effects of anaphthalene acetic acid (NAA) and the combination of NAA and 2,4-D on the culture of the tree peony mature embryo were primarily studied. The suitable NAA concentration and combination of NAA and 2,4-D in the callus induction medium of the mature embryo were defined. The results showed that, the optimization concentration of single NAA was 6.0-8.0 mg/1 (the optimization concentration was 7.0 mg/1 and the highest frequency of callus induction was 71.50%) and the optimization combination was NAA 1.2 mg/1 + 2,4-D 1.0 mg/1 (the frequency of callus induction was 74.32%) in the callus induction medium of the peony mature embryos. The results indicated that too, there was inhibiting effect of high concentration NAA (>1.0 mg/1) on the germination of mature embryos and the morphogenesis. This study has provided a theoretical basis for improving the tissue culture efficiency, establishing the simple and efficient in vitro clonal propagation system, genetic transformation and molecular plant breeding in the tree peony.
