Construction of A cDNA Library and Cloning of MADS-BOX Gene from Dendrobium Nobile
A cDNA library from the flower bud of Dendrobium nobile was constructed. The result showed that the cDNA library contained 3.02×105 pfu, the size of the inserts ranged from 0.5 kb to 2.5 kb, and the percentage of positive clones was almost 100％. The full-length MADS-BOX cDNA was obtained through sequencing clones picked randomly, which suggests some full-ength cDNAs be contained in this library. The MADS-BOX gene cDNA was 808 bp, including 117bp 5 untranslated region and 139 bp of 3 untranslated region; The cDNA had an open reading frame of 552 bp encoded a predicted polypeptide of 184 amino acids involved in the LPPWML motif, which was conserved in all MADSBOX.
