Establishment and Optimization of SRAP Reaction System in Litchi
The impact factors of SRAP-PCR reaction, such as the template DNA amount, Mg2+ concentration, dNTP concentration, primer concentration, Taq DNA polymerase dosage were optimized by step optimization method. The results shew that 25 nL of the reaction system contained Mg2+ 2.0 mmolL, dNTPs 0.2 mmolL1, primer 0.3μmol·L-1, template DNA IS ng, and Taq DNA polymerase 1.50 U. The system was preliminarily carried out in a Fl population with 60 plants derived from MaguilixJiaohesanyuehong, and a clear, reproducible SRAP fingerprinting was obtained, which could be used for subsequent construction of genetic maps and other related research.
