Analysis of RAPD Polymorphism in 15 kinds of Melon Plants

In order to reveal the heredity difference of major genera of the melon plant, by optimilized RAPD technology system and UPGAMA cluster method was used to analyze polymorphism of 15 kinds of plants, which belong to 9 genera. The result indicated that the suitable program of 45 thermal cycles was: denaturing for Imin at 94 V .annealing for lminlOsec at 48 C and primer extension for 2min at 72 C. The PCR reaction system was 20 μ I with 60ng DNA template, 0.4 ft mol primer, 0.3mmol dNTP, 3.0mmol MgClhW Taq and 1 X PCR buffer. 8 screened primers from 100 arbitrary primers could amplify all DNA templates and got 85 bands totally, with each had polymorphism .4 groups were made by clustering, in which Group 1 had ornamental cushaw, Chinese cushaw and pumpkin of cushaw genus, and white gourd; balsam pear, snake gourd and cucumber of other genera; Group Ⅱ included calabash gourd, bottle gourd, watermelon and chayote; Group w had muskmelon, net grain melon and Mapao melon; while dishcloth gourd became 1 group. Classification result of RAPD analysis was identical to plant system classification result ,with heredity similarity of species was higher than that of genera, but heredity difference in cushaw genus was higher than that of other different genera of group I, which was different from the result of plant system classification.
