SYSTEMS BIOLOGY FOR THE RESEARCH OF TCM

multi-component medicines that impact multiple targets simultaneously are better at controlling complex disease systems, are less prone to drug resistance and appealing in many important therapeutic areas. Traditional Chinese Medicine (TCM) is a valuable repository to develop multi-component medicines, well-known for its clinical efficacy, rich experiences and resources, while its development towards modern forms of combination therapeutic drug products requires a major methodological breakthrough. In the research of traditional Chinese medicine (TCM),the problem of fragmentation, such as the separation studies on TCM recipe, syndrome and disease and the independent studies on gene, protein and metabolite, should be treated seriously. Therefore, we propose a novel omics approach, chemomics, in which the phytochemical composition of a herbal formula with demonstrated clinical efficacy is regarded as a global chemome, which can be simplified successively through bioactivity-guided screening to achieve an optimized chemome with minimal phytochemical composition for further drug development, while maintaining its curative effect for a specified indicatioa The chemomics approach is also a necessary supplement to systems biology, providing assays for a complex chemical system. By integrating chemomics with systems biology we generated a system-system approach, Integrative Systems Biology, which may have a great potential for rapid development of modernized new composite medicine. Examples of TCM formulae, Qingkailing, Shuanglongfang, Tangshenfang were presented here to demonstrate the routine for R& D of modernized composite medicines (MCM). With the integration and correlation analysis of information flow of both pharmic chemomes and corresponding biological functions which may be expressed as network of genes, proteins and metabolites, the active fractions and/or effective constituents can be identified and the optional prescription can be developed, and then a new multi-component medicine with definite ingredients and feasible quality control, and the pharmaceutical properties can be evaluated and demonstrated.
