A IMPROVED DUAL-COLOR METHOD FOR THE DETECTION OF SINGLE NUCLEOTIDE POLYMORPHISMS ON MAGNETIC NANOPARTICLES ARRAY
Exploiting the advantages of magnetic nanoparticles (MNPs) array, we developed a new strategy for single nucleotide polymorphisms (SNPs) detection based on dual-color hybridization and magnetic nanoparticles (MNPs) array. Biotinylated single strand PCR products containing the SNP locus were captured onto the MNPs coated with streptavidin. The DNA-MNPs complexes were immobilized on the slide to fabricate MNPs array through magnetic field. The MNPs array was hybridized with the corresponding allele-specific detection probes for each locus. Using MNPs array, all samples can be analyzed in one hybridization chamber which lowers the cost of the assay. One SNP locus from 24 individual samples were discriminated using this platform and the results demonstrated that the expected scores and good discrimination were obtained between the two alleles.
