APTAMER-BASED TURN-ON FLURESCENT ASSAY FOR CELLULAR PRION PROTEIN DETECTION
We have developed a novel turn-on fluorescent assay for the detection of cellular prion protein (PrPc) on the basis of a nucleic acid aptamer and a 3, 4, 9, 10-perylenetetracarboxylic diimide derivative (DAPER). In this PrPc detection system, the strong fluorescence of DAPER is almost completely quenched by the anti-PrPc aptamer (turn-off) j however, in the presence of PrPc, it binds to the DNA aptamer specifically and induces the unrestricted DNA fold into heparin structure, so the DAPER molecules are released that resulting in fluorescence recovery (turn-on).
