EFFECT OF ANTI-TNF-o MAB ON NF-κB NUCLEAR TRANSLOCATION AND PROLIFERATION OF HUMAN HEPG2 CELLS

The activation of NF-κB induced by tumor necrosis factor-α (TNF-α) is closely related to hepatocarcinogenesis and drug tolerance, but the influence of inhibiting NF-κB Activation by TNF-α mAb in hepatoma cells is still poorly understood up to now. In this paper, we investigated the effect of TNF-α mAb on NF-κB activation and nuclear translocation by many methods, such as ELISA, Western Blot, qPCR, immunohistochemistry. The results indicated that the expression level of NF-κB mRNA was significantly inhibited. Besides, NF-κB activation and nuclear translocation were significantly inhibited by TNF-α mAb in HepG2 cells. So it can be seen that intervention with TNF-α mAb is expected to become a new method of HCC targeted therapy.
