CLONING AND CHARACTERIZATION OF A NOVEL METALLOTHIONEIN cDNA FROM KANDELIA CANDEL
A new metallothionein (MT) gene named KMT was cloned from Kandelia candel, a mangrove plant with constitutional tolerance to heavy metals, by rapid amplification of cDNA ends (RACE). The full length of KMT cDNA was 744 bp including 120 bp 5 noncoding domain, 240 bp open reading frame and 384 bp 3 termination. The characters of KMT protein showed it was 79aa in length, the molecular weight was 7. 75 kDa, the pi was 4. 81, and 14 cysteine motifs were detected to distribute at both N-and C-terminals which were thought to be the putative metal irons binding domain. The phylogenetic investigation indicated that the KMT protein sequence shared more than 60% homology with the type 2 MT-like protein genes of other higher plants. At amino acid level, the KMT was shown homology with the MT of Quercus suber (83%), of Ricinns communis (81%) and of Arabidopsis thaliana (64%).
