CLONING OF FULL-LENGTH HUMAN ESTROGEN RECEPTOR AND CONSTRUCTION OF THE EXPRESSION VECTOR IN YEAST
To establish the recombinant yeast estrogen system, cloning and expression of full-length human estrogen receptor(hER) cDNA in yeast The hER cDNA was amplified by RT-PCR method from human breast carcinoma cells,MCF-7, according to the sequence of human full length of estrogen recepor cDNA. The gene was cloned into the yeast expression vector via pPIC9/hER vector, which was transformed into Pichia pastoris. The result showed that recombinant expression vector of full-length human enstrogen recepor cDNA, pPIC9/hER, was constructed successfully, which was the very important groundwork for further establishment of the recombinant yeast estrogen system.
