CONSTRUCTION OF PLASMID CONTAINING HUMAN ARGONAUTE2 GENE
Objective To construct recombinant plasmid for further studying the biological function of the Human Argonautez Gene. Methods The total RNA was extracted from HeLa cells. The cDNA encoding human full-length Argonautez (AGO2) gene was obtained by RT-PCR. The AGO2 gene was subcloned to Fluorescent Protein Expression Vector pKaded-MNl to construct recombinant expression vector named pKaded-MNl/AGO2. The proper cloning was identified by electrophoresis and confirmed by sequencing. Results Restriction endonuclease digestion analysis, sequencing showed that the target gene Argonauts was cloned into pKaded-MN1. Conclusion The recombinant plasmid containing Human Argonautez gene has been achieved successfully.
