DEVELOPMENT AND APPLICATION OF PLUG MARKERS FOR IDENTIFYING THINOPYRUM INTERMEDIUM CHROMOSOME 1ST IN WHEAT BACKGROUND
PCR-based Landmark Unique Gene (PLUG) markers are potentially useful in detecting alien chromatin in wheat backgrounds. In the present study, eight pairs of homologous group-1 PLUG primers were selected to amplify the genomic DNA of Thinopyrum intermedium, Pseudoroegneria spicata and Chinese spring (CS). Eight specific DNA segments were obtained from both Th. intermedium and Ps. spicata, but not in CS. Using a set of wheat-Th. intermedium chromosome addition lines as materials, all PCR fragment were located on 1St chromosome, of which three located on short arm and five located on long arm. Furthermore, wheat-Th. intermedium substitution line 1St(1D) and its cross progeny were scanned using PLUG-PCR, The results indicated that these eight 1-St specific PLUG markers could be used for detecting the transmission and rearrangement of Th. intermedium 1St chromosome in wheat background.
