DOWN-REGULATION OF MICRORNA139-5P IN ESOPHAGEAL SQUANMOUS CELL CARCINOMA
Objectives To explore the expression profile and the potential role of miR-139-5p in esophageal squamous cell carcinoma (ESCC). Methods: Quantitative realtime PCR was used to evaluate the expression level of miR-139-5p in 137 pairs of ESCC tissues and corresponding normal tissues. Genome-wide microarray was carried out on miR-139-5p mimics transferred EC109 cells and control Genes differently expressed by miR-139-5p over-expression were distributed to GO and pathway analysis. Results: miR-139-5p expression level in ESCC tissues was 9. 43-fold lower than adjacent tissues (P<0. 000 1). Totally 74 upregulated (Ratio>2) and 136 down-regulated mRNAs (Ratio<0. 5) were identified as a consequence of miR139-5p over-expression, which were mostly linked to protein phosphorylation, oxidation reduction and biding process, and were involved in several pathways critical related to carcinogenesis, including apoptosis, MAPK and TGF-β signaling pathways. Conclusion: miR-139-5p was significantly down-regulated in ESCC tissues. Over-expression of miR-139-5p in ESCC cell may trigger large amounts of downstream events and reduce the risk of esophageal cancer as tumor-suppressor gene by being involved in apoptosis, MAPK and TGF-β signaling pathways.
