DYNAMICS STUDIES OF EXOGENOUS MIRNA TRANSFECTION BY LIVE-CELL MICROSCOPY AND SINGLE PARTICLE TRACKING

In recent years, microRNAs (miRNAs) have emerged as important gene-regulators and showed great potential in gene therapy. However, the knowledge of their function in vivo is still lacking. In this work, the exogenous miR-21 transfected into HeLa cells was observed for a long time by live-cell microscopy. Their dynamic properties were analyzed by single particle tracking (SPT) and mean square displacement (MSD) calculation. The results showed that the exogenous miRNAs bound to cells quickly and were confined in a small area. Then long time observation and the analysis of motion mode showed they entered cells and changed their motion mode in different intracellular microenvironment.
