HYDROGEL MICROARRAY FOR LOW-COST SNP SCREENING
This paper propoed a hydrogel chip platform to established a low-cost method for screening of SNP. The proposed method based on copolymerization of acrylamideamidemodified PCR products with acrylamideamide monomers and acrylamidemodified slides to prepare gelbased microarray. Then the array was hybridized with two types of hybridization probes for SNP respectively. Electrophoresis was used to remove the nonspecifically bound targets and mismatches. After that, the array was extended with dNTPs containing Cy5-dCTP. Finally, genotyping was based on fluorescence scanning. As the using of general probe during the hybridization, the specific labeled probesis non-essential for each SNPwhich can reduce the cost of screening. 16 samples of OLR1C14417G were detected, the results are consistent with Sequencing. The established hydrogel chip platform is a low-cost method for detection of SNP.
