INFORMATICS AND ALTERATIONS OF HEPATIC GENE EXPRESSION PROFILES DURING HEPATOCYTE MALIGNANT TRANSFORMATION
Molecular mechanisms underlying hepatoccellular carcinoma ( HCC) progression and aggressiveness are still unclear. The hepatocarcinogenesis model was made in SD male rats given foods containing 2fluorenylacetamide. The aberrant liver gene expression in tissues of different stages was analyzed by Affymetrix Rat GeneChip. Genes that were statistically expressed were grouped in categories according to sub-cellular localization, biological process, or molecular function as revealed by the gene ontology biological process description. The list of top hepatocyte transcripts was up-or downregulated in rat hepatomas. Increase and (or) decrease of gene expression can be found during the hepatocarcinogenesis courses. Significant alterations in gene expressions occurred in biological networks, activation of oncogenes, proliferation-related genes, and genes involved in cell-cell communications. HCC gene expression is complexity, and many genes are unknown or have no relationships with hepatocyte malignant transformation as regards in agreement with the multiple genetic events. Indicating that hepatic biology informatics could be useful to monitor hepatocyte malignant transformation.
