MULTIPLEXED LUMINEX XMAP ASSAY FOR DETECTION OF MICRORNAS

The differential expression and the regulatory roles of microRNAs (miRNAs) are being studied intensively these years, which has called for more sensitive, specific and high-throughput methods for detection of short RNA oligonucleotides in a complex mixture of RNA. Here we present a multiplexed and reliable miRNA detection protocol based on Luminex xMAP technology. Currently, assay is designed to simultaneously analyse 15 human miRNAs and an external control, celmir-238 (Caenorhabditis elegans miRNA), which previously has been reported as assay-normalization controls. Results obtained in a 10-fold dilution series assay demonstrated the analytic sensitivity of the present assay was picogram level of total RNA. Overall, the results showed that this multiplexed Luminex xMAP assay could be used for sensitive and high-throughput detection of miRNAs.
