PREPAREATION OF MONOVALENT CIRCULAR SSDNA APTAMER BY ENZYME LIGATION
Nucleic-acid aptamer is a promising therapeutic agent The instability of aptamer in vivo limits its application in clinical therapy, whereas it could be improved by means of modification. Circular aptamer is a promising modification method. Here we described the construction of monovalent circular ssDNA aptamer with ligation primer by T4 DNA Ligase-mediated ligation reaction. Rolling circle amplication (RCA) was used to detect circular aptamer. The obtained circular aptamer was mainly monovalent, possessed nuclease-resistant properties.
