DETERMINATION OF GUANOSINE 3`, 5`-BIS(DIPHOSPHATE) BY CAPILLARY ELECTROPHORESIS WITH UV DETECTION

The intracellular alarmone guanosine 3 ,5-bis (diphosphateKppGpp) is very important in some bacteria and plants due to its key role in the process of governing gene expression by coordinating the cellular responses to environmental stress. In this work, one method is described for the rapid analysis of this highly phosphorylated nucleotide, ppGpp, by capillary electorphoresis with UV detection (CE-UV). Factors affecting separation and quantification such as buffer concentration, buffer pH and buffer additive have been investigated and optimized. The method was validated with respect to the linearity, sensitivity and precision. The analytical time for the detection of ppGpp by CE-UV has been proved to be less than 15min under the optimized conditions(20℃ ,31 cm×50 fim capillary, 50. 0 mmol · L-1 phosphate buffer, 5. 0 mmol · L-1 sodium dodecylbenzenesulfonate, pH 9. 2, applied voltage 15 kV, UV detection at 254 nm). This proposed method was rapid, simple, and effective, and so could be used for the determination of ppGpp.
