Label-Free Amperometric Immunosensor for the Detection of Carbofuran Pesticide
In this study, anti-carbofuran monoclonal antibody was immobilized on the surface of gold elec-trode modified with gold nanoparticle (GNPs) and l-cysteine to construct a label-free amperometric immunosensor for the detection of carbofuran pesticide. Differential pulse voltammograms (DPV) and current response curve were applied to characterize the electrochemical properties of modi-fied process. Effect of antibody immobilization, concentrations of dopamine solution, pH of working buffer and incubation time were studied in detail. Because lcysteine has good electrocatalytic abil-ity on the dopamine, and can amplify current signal, thus dopamine as substrate was used for generating the current signal to detect the immunological reaction. After the immunosensor was incubated with carbofuran solution, the response of the immunosensor to dopamine was obviously inhibited, which leads to a linear decrease in amperometric response of the immunosensor with the increase of carbofuran concentration. Therefore, the proposed immunosensor can be used for the detection of carbofuran pesticide, exhibited good reproducibility, stability and it was suitable for specific detection of pesticide residue.
