Application of Response Surface Methodology in Extraction of Polysaccharides from Auricularia Auricula
A water-soluble polysaccharide was isolated from Auricularia auricula (L.ex Hook.) Underwood and the content of polysaccharides obtained was determined by the phenol-sulfuric acid method.To maximize polysaccharide yield, response surface methodology (RSM) was applied to predict optimum condition for extraction of polysaccharides from Auricularia auricula.A central composite design involving liquid to solid ratio, extracting time and extracting temperature was used, and second-order model for polysaccharide yield was employed to generate the response surface.The optimized condition was as follows: liquid to solid ratio 50.08:1 (mL/g), extracting time 67.54 min and extracting temperature 74.8 C.The predicted the polysaccharide yield at the condition was 17.852 ug/mL.Experimental verification gave the value of 16.961 ug/mL.
