Molecular Cloning and Tissue Expression of the Calpainl(CAPN1) Gene in Goat
Calpain 1 gene is closely related with meat quality in livestock and poultry. Based on (he bovine mRNA sequences (GenBank accession number: AF22119), the cDNA of Calpain 1 was amplified successfully for the first time using T-A clone in Tianfu goat. Results showed that the full-length cDNA of Calpainl was 2151 base pair (bp) encoding a protein with 716 amino acid residues and contained three conserved domains, a catalytic activity region and a catalytic activity centre. Bioinformatic analysis indicated that the secondary structure was mainly alpha helix and random coil contained rich hydrophilic regions, certain phosphorylation loci and protein kinase A/C loci, caprine Calpain 1 was identical to caprine and other speices, especially in mammals. Tissue expression of the gene was all discovered in seven samples. The expression of the 6-month age was the highest in leg muscle. There was a significantly difference between leg muscle, longissimusdorsi and other internal organ tissues.
