The Synergistic Effect of Chrysin and TNFa on Apoptosis in Hep G2 Cells

Chrysin(5,7-dihydroxyflavone) exists widely in plants,honey and propolis. The anticancer property of chrysin has beendemonstrated though the molecular mechanism is not clear. In this study, we found that pretreatment of chrysin could synergistically promote the cell death induced by TNFa in human hepatoma(Hep G2) cell lines according to the morphological changes and appearance of sub-G1 peak. Then we found the synergistic effect of chrysin and TNFa is related to the activation of caspase 8 and caspase 3. In further exploring of mechanism, we found nuclear transcription factorkappa B (NF-κB) activity was inhibited by the combination of chrysn and TNFα. As the result,the expressions of c-FLTPL and cIAP2 enhanced by TNFa were suppressed by the combination,which are antiapoptotic proteins regulated by NF-κB. All Data from this study thus reveal a novel function of chrysin as an anti-cancer agent and enhance the possibility of TNFα in clinical application.
