The Effects of Bisphenol A on Proliferation, Cell Cycle and Apoptosis of HBL-100 Monolayer

Background and aim;To detect the effects of Bisphenol A (BPA) on proliferation,cell cycle and apoptosis of HBL-100 cell line. Materials and methods: E-Screen assay was used to study the effect of E2 and BPA on HBL-100 proliferation and Flow cy-tometry(FCM)was used to study the effect of test materials on cell cycle and apoptosis. Then an estrogen receptor(ER) inhibitor (ICI182780) was added into the system to detect the changing on the effects of test materials. Results :The highest cell proliferation rate of BPA was at 1.0×10-6mol/L and by this dose BPA promoted more cells entering G2/M phase of cell cycle. All the promoting effects of BPA were higher the E2 s. After adding ICI182780 into the system, the promoting effects on proliferation and cell cycle changing of E2 were vanished when the effects of BPA decreased but still have statistical significance compared with solvent control (P < 0. 05). Conclusion;BPA has obvious estrogen-like activity. It can simulate HBL-100 DNA replication and cell division through ER pathway. BPA may have other pathways through which it can exert simulation effects on cell proliferation and cell cycle alteration.
