Chromosome Aberration Test of Chloroacetophenone (CN) in Chinese Hamster Lung Fibroblasts (CHL)

The mutagenic effects of CN were evaluated by chromosome aberration test of CN in CHL. IC50 value for CN in CHL was 1.42 μg/ml. Five doses (1.50, 0.75, 0.38,0.19, 0.09 μg/Ml)were determined according to the IC50 value. At the same time, the controls of blank, solvent (DMSO) and positivity (mitomycin C and cytoxan) groups were tested. Every test included two sets of experimental groups with and without S9 metabolic activating system. Each dose was composed of triplicate. Results indicated that the chromosome aberration frequencies in different concentrations of CN were 1-4% under the condition with or without S9 metabolic activating system. The chromosome aberration frequencies of the blank and the solvent control were ≤1%. No significant difference (P>0.05) was observed in chromosome aberration frequencies of CHL between the treatment groups and controls of blank and solvent. But the chromosome aberration frequencies of two kinds of positive controls were 24-29%. Which showed that mitomycin C and cytoxan possessed mutagenic effects. So, chromosome aberration effect of CN was negative.
