Subchronic Toxicity Evaluation of Diphenylchloroarsine in Rats

The groups of 20 SD rats of each sex were gavaged with 0, 0.188, 0.377, 0.754 and 1.508 mg/kg of DA in sesame oil 6 d/wk for 13 weeks. The results indicated that after 3 weeks of exposure, salivation and rhinorrhagia occurred in the partial rats in 1.508, 0.754 and 0.377 mg/kg doses. During the exposure, the mean food intakes per 100 g body weight and the increasing rates of the body weights of the rats in 1.508 mg/kg were obviously diminished. The changes of the hematological data occurred in the rats exposed to 1.508 mg/kg of DA. The red-cell counts and the contents of haematoglobin, hematocrits and lymphocytes were obviously reduced, and the content of neutrophil was significantly increased. The analytic results of the blood biochemical data indicated that the concentrations of potassium, total protein, total bilirubin, cholesterine and the activity of aspartate aminotransferase were increased in 1.508 mg/kg. In 0.754 mg/kg, the activity of aspartate aminotransferase of the rats was obviously increased too, and it was not recovered to the normal level at 17 weeks. Histopathologic examinations revealed that the hyperplasia of the tubular structures in the portal area of the livers, the focal necroses of the hepatocytes, the vacuolar degeneration of the hepatocytes and the inflammatory cell infiltration were presented in the rats in 1.508 and 0.754 mg/kg. In addition, the toxicities of 1.508 and 0.754 mg/kg of DA on gastric mucosa, digestive tract, kidney, and adrenal cortex, etc were observed. 1.508, 0.754 and 0.377 mg/kg of DA could cause and enlarge the chronic inflammatory foci of the lung. No abnormal phenomena were found in 0.188 mg/kg. We considered that the main target organs of the toxic effects of DA on the rats (ig) were liver, stomach, kidney, and it could cause and extend the chronic inflammatory foci of the lung. The estimated dose for NOAEL for DA in rats was 0.188 mg/kg when administered orally (ig) for 13 weeks.
