Spatio-temporal Variations of Microcystis Genotypes in Lake Taihu
The structure of gvpA-gvpC gene cluster that encodes the gas vesicle proteins is highly variable in Microcystis. Compared to rDNA, phycocyanin (cpc), rbcLX genes or the intergenic regions, the gvp gene cluster or the gvpA-C intergenic region shows much higher diversities. In the present study, we employed gvpA-C intergenetic sequences (gvpAC-IGS) as molecular makers to investigate the annual spatio-temporal variations of Microcystis in Lake Taihu. Microcystis samples were collected from twenty-two locations in the lake from Oct., 2008 to Dec., 2009. The gvpAC-IGS were generated by PCR and cloned into a T-vector in E. coli. A library of gvpAC-IGS were constructed for each month. From the sequences of 100 clones, we calculated the percentage of each type of gvpAC-IGS. More than 20 types of sequences were identified, of which type 1 was dominating (> 50%) in most months. From Sept. to Nov. in 2009, however, type 1 was greatly decreased in the population, while the diversity of gvpAC-IGS was remarkably increased. Using the same method, we also evaluated if there was any difference of the Microcystis population between different regions or locations in Lake Taihu. On the other hand, we isolated Microcystis clones of different gvpAC-IGS genotypes from the lake. With these isolates, it may be possible to reconstruct the dynamics of population succession under simulating conditions.
