Hydrogen Peroxide Induces Programmed Cell Death in Microcystis aeruginosa in a Dose-dependent Manner

To understand whether Microcystis aeruginosa could execute programmed cell death (PCD), we investigated the morphological, molecular and physiological features of M. aeruginosa with H2O2 treatment, a traditional PCD inducer. We found the growth of M. aeruginosa was weakly affected with 150 μM H2O2 treatment, but dramatically decreased with 250 and 325 μM H2O2 treatment. From the TEM image, plasma membrane blebbing, ambiguous nucleoid zone, deformation, partial disintegration of thylakoids and cytoplasmic vacuolation, intact plasma membranes were observed, which were similar features to that of PCD in eukaryotes. Correspondingly, Hoechst 33342 staining showed highly condensed nucleoid chromatin in the central region of cells, and a loss of nucleoid chromatin architecture or empty cells with complete lysed nucleoid. Moreover, cellular degradation was concomitant with dramatic decreases in photosynthetic efficiency (Fv/Fm, ETRmax).
