Ultrastructural effects on hepatopancreas of Bellamya aeruginosa (Gastropoda) fed with toxic cyanobacterial suspension and recovery
Cyanobacteria can produce hepatotoxic microcystins (MCs) which is harmful to aquatic animals and poses a threat to human health. The purpose of this study was to evaluate the toxic effects of Microcystis aeruginosa on Bellamya aeruginosa via investigation of ultrastructural alterations in hepatopancreas after exposed to the toxic cyanobacteria to further understanding the hepatic intoxication and detoxification mechanism in snail. B. aeruginosa were exposed to in both treatments (Cyanobacteria only group and mixed with green alga group) for 15 days. Intoxication was followed by 15-day detoxification period. The MCs concentration of the algal suspension and the kinetics of MCs accumulation and depuration in the gastropods were established using the ELISA. Total MCs concentration was 18.67±2.80μg L-1 in the mixed suspension, 37.93±13.85μg L-1 in the cyanobacterial suspension and undetected in the control suspension. MCs concentration was detected in the hepatopancreas of the two experimental groups and increased with time during the intoxication period. After the removal of the toxic cyanobacterium, MCs concentration in the hepatopancreas was depurated rapidly. Ultrastructural changes in the hepatopancreas were investigated using the transmission electron microscope technique. Severe cytoplasmic vacuolization, increases in mitochondria, reduction and disruption of microvilli and apoptotic body were observed. These alterations included bizarre nuclei that ranged in their degenerative changes from severe karyorrhexis to complete pyknosis. All cell organelles began recovery after the snails were transferred to the non-toxic Scendesmus quadricauda. Our results suggested that apoptosis of hepatocytes induced by MCs may be an important mechanism of hepatotoxicity to B. aeruginosa.
