Genetic Diversity of Cyanobacterial Blooms in a Tropical Reservoir Using the 16S-23S rRNA Internal Transcribed Spacer and mcyH gene

A study was conducted to examine the dynamics and genetic variability of Microcystis in a tropical eutrophic reservoir in Singapore from February 2008 to August 2009. The abundance of the Microcystis population and percentage of toxigenic cells were determined by real-time PCR (qPCR) assays targeted at 16S rRNA and mcyE genes. Genetic diversity of cyanobacterial community and Microcystis species were analyzed based on the 16S-23S rRNA internal transcribed spacer (ITS). Clustering of the sequences show that Microcystis, Anabaena/Aphanizomenon, Synechococcus and Oscillatoria were genera in the reservoir. The Microcystis ITS phylogenetic trewe ecaren tbhee d divoimdeidn ainntto 4 clusters where 50% of the sequenced clones were grouped in Cluster II, which was shown to contain only microcystin producers. Temporal changes of ITS genotypes were observed but there was no spatial genetic difference between samples.
