Genetic variation patterns of Medicago ruthinica populations from north China
Genetic diversity was investigated for twenty four populations of Medicago ruthenka from North China using allozyme and microsatellites markers. We analyzed the genetic variability using six allozymes loci and five polymorphic microsatellite loci. The two assays differed in the amounts of polymorphism. Micro -satellites detected the higher amounts of polymorphism. For allozymes, genetic diversity between population(Hs) averaged 0.2733 and within population genetic diversity(DST) averaged 0. 0473, mean coefficient of gene differentiation for loci (GST) in the populations was estimated 0. 1813. As for gene flow at populations level, the gene flow according to GST was 1.4427 and 1.2040 for allymes and SSR markers respectively, which indicated there were definite gene flow between populations. Low differentiations among populations at allozymes (FST=0. 1719) and SSR markers (FST=0. 1816) were found. This study showed there were high genetic diversity in M. Ruthenka populations of North China both using allozyme and SSR markers.
