Degradation of hemicelluloses of cottonseed coat by xylanase

The hemicelluloses were separated from the cottonseedcoat and degraded with Pulpzyme HC. The digestion degreevalue was calculated by the equation depended on the release ofreducing sugars concentration which was determined from DNSfitting curve equation. The optimum enzyme reactiontemperature was 60 ℃and pH was 5.5. The duration of 80 minwas chose as the most suitable enzymolysis time. Thedegradation degree value was 57.5% when 0.24 ml purePulpzyme HC was added with one milligramme hemicellulosespowder. The K+content substrate (0.5 mmol L -1) enhanced theenzymic degradation of hemicelluloses. The other metal ionswere found to be some inhibitors of Pulpzyme HC. The effects ofchelating agent (EDTA) were complex and no trend wasobserved.
