Extraction of mycelial polysaccharides from submerged-cultured Agaricus blazei
Aqueous extraction and alkaline (NaOH) hydrolysismethod were comparatively studied in polysaccharides extractionfrom submerged-cultured Agaricus blazei mycelia. The resultsshow that the polysaccharide content and the total extract ratefrom alkaline extraction method were obviously higher thanthose from the aqueous extraction method. Optimization resultsby the orthogonal matrix method on polysaccharide extractionfrom A. blazei show that the optimum alkaline concentration was1.0 mol/L, hydrolysis temperature 60 ℃, hydrolysis time 1 hour,and liquid: solid ratio 15. With the optimized condition, thepolysaccharide extraction rate reached 11.93 ± 0.68 %. Themycelial polysaccharide obtained with alkaline hydrolysismethod was fractionated by the DEAE column chromatography.Five APS fractions were isolated in the polysaccharide samplewith APS-1 as the main component.
