Lamellipodium Locomition of the Daughter Cells Facilitates the Cytokinesis of NRK Cells without Myosin II

Cytokinesis B serves as a backup mechanism in somemammalian cells after cytokinesis A failure. However, itsmechanism remained poorly understood. Here, we appliedcytochalasin D, an known actin inhibitor, globally duringcytokinesis of NRK cells or cells dividing using cytokinesis B.Different from large fan-shaped lamellipodia at the leading edgeduring cytokinesis B, the progression of cytokinesis B wasobvious inhibited in presence of 1 μ M cytochalasin D. Pole to poledistance and cellular apparent surface area of the two daughtercells during cytokinesis B decreased significantly in presence of1 μ M cytochalasin D. Immunofluorescence staining revealed thata mass of actin was found to recruit from the cytoplasm to thefan-shaped leading lamellipodia of daughter cells in cells treatedwith blebbistatin. Our results indicated that lamellipodiumlocomition of the daughter cells mediated by actin whichgenerated active traction force facilitated the progression ofcytokinesis B.
