Study on Toxicity Action of Nonylphenol on Human Embryo Liver L-02 Cells

Objective: Nonylphenol(NP) is NPnEOs of recalcitrantcompound and classified as an endocrine disruptingchemical(EDC). The reproductive toxicity of Nonylphenol hasbeen extensively studied in mammals. In this study, To explorethe toxic effects of nonylphenol on human embryo Liver L-02Cells.Methods Human embryo Liver L-02 Cells were exposed todifferent concentration of NP(1 、10 、50 、100 μmol/L).Survivalrate was measured by MTT, apoptosis rate was measured by flowcytometry, level of Superoxide Dismutase (SOD), MaleicDialdehyde(MDA), Glutathione Peroxidase(GSH-PX) weremeasured by Typical reagent kits.Rusults L-02 Cells survival ratedecreased as the concentrationof NP increased. and prolongationof culture time (50 μmol/L NP for 0, 1h, 4h, 24h) decreased.Apoptosis rate increased as the NP concentration ,Comparedwith the normal control group, the MDA content of the middleand high dose of NP group increased obviously (p<0.05). TheGSH-PX activity of high dose of NP group was also higher thanthe normal control group(p<0.05). The SOD activity of all dose ofNP groups were all obviously lower than the normal controlgroup(p<0.05). Three kinds of measurement of the positivecontrol group were all significantly change than the normalcontrol group (p<0.05).Conclusion NP has cytotoxic effect andoxidative damage to L-02 Cells, and induce L-02 Cells apoptosis.
