The Contribution of Polar Cortical actin and Microtubule to Cytokinesis at Early Anaphase
Temporally and spatially regulating the microtubulecytoskeleton, the actin cytoskeleton and cortical myosin assemblyis primary to cell dividing. To realize the contribution of actinand microtubule in polar cortex at early anaphase, we locallyreleased cytochalasin D and colchicine in a highly localizedmanner to polar region of dividing normal rat kidney cells. Theresults showed that local release of CD or colchicine at the polarregion caused inhibition of cytokinesis at early anphase beforeingression. Cells treated with 30 μ M blebbistatin were used tocontrast to control. Our findings showed that disturbed polarcortical cytoskeleton not only inhibited initiation of cytokinesisbut also influenced the position of furrow. The results suggestedthe definitive roles of global coordination of cortical activities atearly anaphase.
