A Novel Method using Morphological Gradient for Spot Extraction from cDNA Microarray Image
Gene expression analyses by probes of hybridizationfrom mRNA to cDNA targets arrayed on membranes or activatedglass surfaces have revolutionized the way of profiling mega levelgene expression. The main remaining problems however aresensitivity of detection, reproducibility and data processing.During processing of microarray images, especially irregularitiesof spot position and shape could generate significant errors. Herewe report a novel method to eliminate such obstacles by sensingtheir edges. Application of morphological gradient technology onseparating spots from the background decreases the probability of the errors and gives more accurate information about the states ofspots such as the pixel number, degree of fragmentation, width andheight of spot, and circumference of spot. Such information can beused for the quality control of cDNA microarray experiments andfiltering of low quality spots. In this paper we proposed a methodwhich performing accurate spot segmentation of a microarrayimage, using morphological gradient image analysis techniquesand compared the result with that of conventional methods, whichincluding fixed centre method (using ScanAlyze) and unfixedcentre method (using GenePix). The result of the experimentshows that the method presented in this paper is accurate,automatic and robust.
