The effect of Sargassum fusiforme polysaccharides on the functioning of erythrocyte membrane of S180 mice by affecting Ca 2+ channels
To analyze the effect of Sargassum fusiformepolysaccharides (SFPS) on the functioning of erythrocytemembrane of S180 mice by affecting Ca 2+channels. A tumormouse model was developed, and the mice were treated with high,medium, and low doses of SFPS. Changes in Ca 2+ in theerythrocytes in the tumor-bearing mice were measured using laserconfocal scanning; changes in the content of sialic acid and theacitivities of ATPase were meausred using relevant test kits;changes in the level of erythrocyte membrane potential wereanalyzed using FCM; and changes in the complex mobility oferythrocytes were measured using HPCE. SFPS lowered theintracelluar Ca 2+ of erythrocytes in tumor-bearing mice,increased the content of sialic acid on surface of erythrocytemembrane, increased the acitivities of Na+,K+-ATPase and Ca 2+,Mg 2+-ATPase, and raised the level of erythrocyte membranepotential and the complex mobility of erythrocytes inelelctrophoresis. By affecting the Ca 2+channels, SFPS canincrease the content of sialic acid on the surface of erythrocytemembrane. The sialic acid combines with Ca 2+to increase theactivities of Na+,K+-ATPase and Ca 2+, Mg 2+-ATPase, resulting in arise in the membrane potential of the erythrocytes and an increasein the complex mobility of the erythrocytes in electrophoresis, thusrestoring many physiological and biochemical functions of theerythrocyte membrane. This may be one of the anti-tumormechanisms of SFPS.
