Effects of juglone on ROS production and mitochondrial transmembrane potential in SGC-7901 cells
To investigate the effects of juglone on ROSproduction and mitochondrial transmembrane potential(ΔΨ m) in SGC-7901 cells in vitro, further to elucidate theapoptosis mechanism. The apoptosis morphological changesof SGC-7901 cells were observed using fluorescencemicroscope. The level of ROS and ΔΨ m of cells were detectedby FCM. The change of Ca 2+I in cells stained withFluo-3/AM was observed by LCSM. Hoechst 33258 stainingassay showed thick granulated fluorescence in condensednuclei and cytoplasm, and the number of apoptotic bodieswas increased with increasing of juglone concentration. Thelevel of ROS at the dose of 5, 10, 15 and 20?M of juglone was34.83±1.45%, 42.43±1.36%, 53.73±1.38% and 68.67±1.33%,respectively, which were significantly higher than that incontrol group (P<0.01). The concentration of Ca 2+in cellsexposed to juglone for 24 h was increased significantly in adose dependent manner compared with control group(P<0.01). ΔΨ m of treatment groups was 85.53±1.82%,53.57±2.48%, 46.33±1.46%, 36.43±2.64%, respectively, whichwere significantly lower than that in control group(P<0.01).Therefore, juglone can increase intracellular level ofROS and concentration of Ca 2+, decrease ΔΨ m and induce thecells apoptosis.
