Curcumin upregulate HO-1 expression but downregulate HO-2 expression in SHSY5Y cell
Curcumin ,the most active constituent ofturmeric ,is currently one of the major anticancenew drugs and has been reported to enhance HemeOxygenase-1(HO-1) in human brain whenoxidative stress occur. Keeping the balance ofHO-1 and HO-2 plays an important role in thebrain. However, few researches study the effects ofcurcumin on the two isoenzymes of HO. Here weinvestigated the effects of curcumin on the changeof HO-1 and HO-2 in neuroblastoma cell lineSHSY5Y. We found that the mRNA and proteinlevels of HO-1 were increased in a dose-andtime-dependent manner (P<0.05) in the cells aftertreated with Curcumin, whereas, the expression ofHO-2 were gradually downregulated in a dose-andtime-dependent manner (P<0.05). A negativecorrelation was found between HO-1 and HO-2.These results indicate that Curcumin regulatedHO-1 and HO-2 bilaterally, which may have animportant effect on the pathogenesis anddevelopment of carcinoma, and may provide a newdirection for disposing of carcinoma.
