Phosphatase Activity Correlation with pH in Landfill and Optimization of the Method for Determining
The paper aims at exploring the relation between phosphatase activity and pH in landfill, optimizing the experiment condition of phosphatase in landfill on the basis of the soil enzymology and studying the characteristics of three kinds of phosphatase under different stages of refuses decomposition. The results show that the activities of Acidic Phosphatase (ACP)and Alkaline Phosphatase (AKP) activity were closely related to the pH of wastes in landfill, the pH value in landfill were measured to determine which one to be used to present the total phosphatase (TPase) activity. The optimal experimental conditions under which the activities of ACP were tested were 30 ℃ for dried temperature, 0.3ml methylbenzene and 25ml of 0.5% disodium pheny phosphate and 4.0ml acetate buffer (pH5.0) respectively, Then the culture temperature was 45 ℃and culture period lasted for 6h. The activities of Neutral Phosphatase(NEP) were measured on the optimal condition of 30 ℃ of dried temperature, 0.4ml methylbenzene and 25ml of 0.5% disodium pheny phosphate and 3.0ml citric acid buffer (pH7.0), culture temperature were 45 ℃ and 3h. As for AKP, the activities were measured at 30 ℃. The dosages of methylbenzene, disodium pheny phosphate (0.5%) and NH4CL-NH4OH buffer (pH9.8) were 0.4mL, 25mL and 3.0mL respectively, Culture temperature and culture period were 50 ℃ and 24h. The Relatively Standard Deviation(RSD) of means of ACP, NEP and AKP was lower than 1.9%, 1.7%, 1.6% respectively. The results were reliable and high-precision.
