Affinity membrane chromatography: bromelain adsorption using Remazol Brilliant Voilet 5R as a ligand

A composite membrane was prepared by covalent coupling chitosan (CS) to activated nylon membrane, which greatly decreased the non-specific adsorption of protein. The affinity dye, Remazol Brilliant Voilet 5R was immobilized onto the CS-coated nylon membrane. In this paper, the binding characteristics of bromelain to the dye-ligand immobilized membranes have been studied from aqueous solution in a batch system. The loading of Voilet 5R on the CS-coated nylon membrane was 102.5 μmol/g membranes. The adsorption capacities of dye-ligand affinity membrane reached the max at about pH 8, but decreased with increasing NaCl concentration. The Voilet 5R immobilized membrane provided an efficient way of bromelain purification from aqueous solution, showing high adsorption capacity and high selectivity for bromelain. We also performed the dynamic experiment for Voilet 5R, which indicated a high bromelain adsorption and desorption rate.
