Preparation of bacterial cellulose pellets as carriers for laccase immobilization
Bacterial cellulose (BC) is pure cellulose synthesized by some bacteria. However, the application of BC pellets in enzyme immobilization was rarely reported. The form of BC is usually membrane prepared by static cultivation traditionally. In the present research, a kind of bacterial cellulose beads,the particle size of which ranges from 8 to 10 mm, were prepared. After alkali treatment and freeze-drying, the pellets were used for laccase immobilization by adsorption. The properties of immobilized enzyme were characterized. The results showed that the immobilized laccase exhibited the maximal activity at pH 3.5 and the reaction temperature was 60℃. Compared with free enzyme, the pH and thermal stabilities of the enzyme were increased after the immobilization. Therefore, bacterial cellulose pellets are practical and potential biomaterial carriers in enzyme engineering.
