The Chemical Conposition and Extraction Process of Ginkgo Protein
Abstract: Kjeldahl method was used to measure the protein content from ginkgo seeds in different area,i.e.Jiangsu, Hubei and Guangxi in china, and in different species,i.e., Longyan ginkgo,Dafuzhi ginkgo, round-shape ginkgo, potato-shape ginkgo, plum-shape ginkgo, bergamot ginkgo. Composition of ginkgo protein and its extraction process were also studied in this paper.The results showsed that protein contents in different species were at 7.86%~11.88%, moisture at 56.01%~60.01%, the relative protein contents in five extract parts,i.e.,water-soluble, salt-soluble, alcohol-soluble,alkali-soluble and insoluble,were 22.20%, 17.99%, 1.73%, 30.32% and 25.97% respectively. The highest extract yield of salt-soluble protein was 99.516% by ultrasound-assisted extraction at the optimal conditions of 0.14mol/L NaCl solution as extraction agent,PH8, solid-liquid ratio 1:7 (m/v), extraction 30min, 3 times. By using DEAE-cellulose, we can get salt-soluble protein at a purity of 90.77%, which was further treated by HCl for analyzing on changes of amino acids by HPLC,the result demonstrated Lysine climbed from 1.25% to 4.53%, Serine raised from 1.83% to 10.55%, and Histidine 10.34% to 44.24%.
